Isso pode ser devido \u00e0 falta de sensibilidade de alguns dos m\u00e9todos de detec\u00e7\u00e3o e ao fato de que alguns v\u00edrus respirat\u00f3rios n\u00e3o serem sistematicamente procurados. O objetivo deste teste \u00e9 usar m\u00e9todo molecular r\u00e1pido, sens\u00edvel e espec\u00edfico para a detec\u00e7\u00e3o de um grande painel de v\u00edrus de RNA\/DNA respirat\u00f3rio que s\u00e3o mais poderosos do que os m\u00e9todos cl\u00e1ssico de ensaio de imunofluoresc\u00eancia e cultura. Consideramos procurar adenov\u00edrus, frequente causador de infec\u00e7\u00f5es do trato respirat\u00f3rio, em um ensaio de PCR multiplex incluindo outros pat\u00f3genos respirat\u00f3rios de DNA.<\/p><p>Utilizamos o m\u00e9todo primer-sonda para detec\u00e7\u00f5es de fragmentos do DNA\/RNA virais em equipamentos de PCR em tempo real (RT-qPCR ou transcriptase reversa qualitativa e quantitativa PCR). Logo, este m\u00e9todo de detec\u00e7\u00e3o \u00e9 direto. A tabela 1 apresenta as informa\u00e7\u00f5es dos protocolos de amplifica\u00e7\u00e3o utilizados assim como a compara\u00e7\u00e3o deles com alguns kits comerciais.<\/p><p>INTERPRETA\u00c7\u00d5ES<\/p><p>Os v\u00edrus e respectivas regi\u00f5es-alvos de seus genomas est\u00e3o descritos na primeira e segunda colunas, respectivamente. Os limites de detec\u00e7\u00e3o apresentados na terceira coluna foram estimados atrav\u00e9s da dilui\u00e7\u00e3o seriada dos genomas virais, sendo consideradas os menores n\u00fameros de c\u00f3pias para os quais 100% das rea\u00e7\u00f5es foram positivas. As porcentagens de concord\u00e2ncias dos resultados positivos e negativos obtidos com nosso protocolo e outros protocolos comerciais est\u00e3o apresentados na quarta e na quinta colunas, respectivamente.<\/p><p>Tabela 1.
